棉花CBF2基因克隆和超表达CBF2棉花增强抗冷性

摘 要:

CBF (C-repeat binding factor)是一类植物特有的转录因子, 其主要功能是提高植物的抗冷性。本研究从陆地棉(Gossypium hirsutum) ‘柯字棉312’中克隆到1个棉花CBF基因, 命名为GhCBF2。蛋白序列分析结果表明, GhCBF2与其他植物的CBF蛋白类似, 含有一个由216个氨基酸组成的AP2转录因子保守结构域。在棉花幼苗受到冷处理后, GhCBF2基因的表达水平提高。在冷胁迫处理后, 过量表达GhCBF2基因棉花的存活率要显著高于野生型棉花, 其细胞膜的相对渗透率显著低于野生型棉花, 而可溶性糖含量显著提高。以上研究表明, Gh-CBF2基因可能在提高棉花抗冷性发挥作用。

Overexpression of cotton CBF2 gene enhances cold tolerance in transgenic Gossypium hirsutum

Abstract:

CBF/DREB proteins are transcription factors in plants, which play important biological functions in the cold stress resistance. In this study, a CBF/DREB gene, named GhCBF2, was cloned from Gossypium hirsutum. The sequence analysis indicated that GhCBF2 coded a protein consisted of 216 amino acids and contained a typical AP2 conservative domain, which was similar to CBF proteins in other plant species. The expression level of GhCBF2 gene was increased after cotton seedlings were cold treated. The survival rate of overexpressing GhCBF2 cotton was significantly higher than that of wild type cotton. Meanwhile, the soluble sugar content in the leaves of transgenic plants was higher than wild type cotton, while the cell electrolyte leakage was lower than wild type cotton. The above results showed that GhCBF2 may enhance the transgenic plants tolerance to cold stress.
